Located at 250 East 25th Street, at the southwest corner of Second Avenue, the ground-up building takes its name from Hendrick Kip, a Dutch colonist who arrived in New Amsterdam in 1637 and settled along the East River on an inlet that would eventually be named Kips Bay. Over a century later, a property that once belonged to Hendrick’s brother Jacobus was leased as a quarantine facility for patients with yellow fever. The property later evolved into Bellevue Hospital, one of the medical institutions Kips Bay is well known for in the present day.

Hendrix House is strategically positioned near the proposed Science Park and Research Campus (SPARC) Kips Bay, a $1.6 billion life sciences hub envisioned by the city and state government. The construction contract was announced in September 2024, and the first of two phases of construction is expected to begin in late 2025.

In addition to the hospitals and forthcoming life sciences hub, Hendrix House benefits from close proximity to Baruch College, Madison Square Park, critically acclaimed Gramercy restaurants, supermarkets like Fairway and Trader Joe's, and a robust nightlife scene on Third Avenue. Transportation options include the 23rd Street 6 train and the East 34th Street ferry terminal.

Kips Bay has also attracted attention in recent years amidst a growing scarcity of development sites in adjacent neighborhoods. In January 2023, Manhattan Borough President Mark Levine published a report envisioning rezoning the west side of Second Avenue between East 23rd and East 34th streets, potentially adding up to 942 residential units.
